Biological Background: APAF1 Function and Localization
- APAF1 encodes the apoptotic protease-activating factor 1 (Apaf-1), a cytoplasmic protein that plays a central role in the intrinsic apoptosis pathway.
- Upon cytochrome c release from mitochondria, Apaf-1 oligomerizes and, in an ATP-dependent manner, recruits and activates pro-caspase-9 (Apaf-3), triggering downstream caspase-3 activation and apoptosis.
- Isoform 6 of APAF1 is known to be less effective in inducing apoptosis compared to the canonical isoform.
- APAF1 is ubiquitously expressed, with highest mRNA levels in adult spleen and peripheral blood leukocytes, and in fetal brain, kidney, and lung. Isoform 1 is detected in heart, kidney, and liver.
- APAF1 is classified under keywords including apoptosis, ATP-binding, nucleotide-binding, calcium, WD repeat, and alternative splicing, reflecting its functional and structural complexity.
Experimental Guidance and Technical Tips
- The immunogen used for this antibody corresponds to amino acids 470–600 of human APAF1 (NP_863651.1), a region that may be conserved across isoforms; consider verifying isoform specificity in your system.
- For Western blot (WB), multiple bands may be observed due to APAF1 isoforms and potential post-translational modifications; the predicted molecular weight range is 38–142 kDa. Validate band identification using appropriate controls.
- This polyclonal antibody is suitable for WB, IF/ICC, and ELISA applications as indicated; for each assay, optimize antibody dilution and detection conditions in the relevant sample matrix.
- Given the cross-reactivity with human, mouse, and rat, this antibody can be used for comparative studies across these species; confirm cross-reactivity in your experimental setup.
