Biological Background: ANTXR2 Function and Localization
- Protein identity: ANTXR2 (Anthrax toxin receptor 2) is also known as capillary morphogenesis gene 2 protein (CMG-2). It is a single-pass type I membrane protein with a von Willebrand factor A (vWA) domain and is expressed in multiple isoforms due to alternative splicing.
- Extracellular matrix interactions: Necessary for cellular interactions with laminin and the extracellular matrix, playing a role in cell adhesion and basement membrane assembly.
- Anthrax toxin receptor – binding: Acts as a receptor for the protective antigen (PA) of Bacillus anthracis. Binding of PA triggers heptamerization of the receptor-PA complex. Related references: PMID:12700348 PMID:15243628 PMID:15326297
- Anthrax toxin receptor – internalization: Following PA binding, the complex relocates to glycosphingolipid-rich lipid rafts and is internalized via clathrin-dependent endocytosis. In the acidic endosomal environment (pH < 7), the complex rearranges to form a pore, allowing anthrax lethal factor (LF) and edema factor (EF) to translocate into the cytoplasm. - Subcellular localization: Found at the cell membrane, endoplasmic reticulum membrane, and also secreted. The secreted form may arise from alternative splicing or shedding.
- Tissue distribution: Widely expressed, with highest levels in prostate, thymus, ovary, testis, pancreas, colon, heart, kidney, lung, liver, peripheral blood leukocytes, placenta, skeletal muscle, small intestine, and spleen.
- Post‑translational modifications and features: Glycoprotein with disulfide bonds; also classified as a phosphoprotein and metal‑binding protein. Disease variants are associated with hyaline fibromatosis syndrome and infantile systemic hyalinosis.
Experimental Guidance and Technical Tips
- The immunogen (aa 34–300) spans the extracellular vWA domain, which is predicted to be exposed on the cell surface and accessible in fixed/permeabilized preparations.
- For Western blotting, the theoretical molecular weight is ~54 kDa, but glycosylation and isoform-specific variants may produce additional bands; always validate in the relevant sample system.
- For IHC-P, antigen retrieval and fixation conditions should be optimized; tissues with high ANTXR2 expression (e.g., prostate, colon) serve as positive controls.
- In IF/ICC, permeabilization is required to detect intracellular (ER) pools; cell lines expressing ANTXR2 (e.g., some prostate or colon cancer lines) can be used for validation.
- As a polyclonal antibody, each lot may exhibit slight variations; it is advisable to titrate the antibody and confirm specificity with appropriate blocking peptides or knockout controls where possible.
