Biological Background: ZWINT Function and Localization
- ZWINT, also known as SIP30, ZWINT1, KNTC2AP, HZwint-1, is a component of the outer kinetochore KNL1 complex that serves as a docking point for spindle assembly checkpoint components and mediates microtubule-kinetochore interactions. Related references: PMID:15094189 PMID:15485811 PMID:15824131
- It is a subunit of the KMN network (MIS12, NDC80, KNL1 complexes) that constitutes the outer kinetochore, connecting centromeric DNA to spindle microtubules and recruiting mitotic checkpoint proteins. Related references: PMID:15094189 PMID:15485811 PMID:15824131
- ZWINT targets the RZZ complex to the kinetochore at prometaphase (PubMed:15485811) and recruits MAD2L1 (By similarity), contributing to spindle assembly checkpoint activation. Related references: PMID:15485811
- Beyond mitosis, ZWINT is essential for alignment of homologous chromosomes during meiotic metaphase I and activates the spindle assembly checkpoint at unattached kinetochores in meiosis I. Related references: PMID:15485811
- Subcellularly, ZWINT localizes to the nucleus, specifically at the centromere-kinetochore region of chromosomes, consistent with its role in chromosome segregation.
- The protein contains a coiled coil domain, is subject to alternative splicing, and its 3D structure has been partially resolved (UniProt Keywords).
- As a key kinetochore component, ZWINT is implicated in cell cycle regulation, mitosis, and chromosome biology, underscored by its identification in proteomics studies.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant fusion protein covering amino acids 1–277 of human ZWINT (NP_008988.2), corresponding to the N-terminal region that includes functional domains. Consider using this antibody to detect full-length and alternatively spliced isoforms.
- For Western blotting, the predicted molecular weight is ~31 kDa. Include appropriate positive control lysates from mitotic cells or tissues expressing ZWINT, and validate the band size in your sample system.
- In immunofluorescence/ICC, expect punctate kinetochore staining during prometaphase and metaphase in dividing cells. Counterstain DNA to visualize chromosomes and confirm localization.
- For ELISA, optimize coating antigen and antibody concentrations using recombinant ZWINT protein or cell lysates as standard.
- Cross-reactivity with human, mouse, and rat has been reported; however, validate in the specific species and experimental context to ensure specificity.
