Biological Background: ZRANB3 Function and Localization

  • ZRANB3 (DNA annealing helicase and endonuclease ZRANB3), also known as AH2 (annealing helicase 2) or zinc finger Ran-binding domain-containing protein 3, is a multifunctional enzyme that maintains genome stability at stalled or collapsed replication forks by facilitating fork restart and limiting inappropriate recombination during template switching events. Related references: PMID:21078962 PMID:22704558 PMID:22705370
  • Recruited to sites of stalled DNA replication by polyubiquitinated PCNA, ZRANB3 acts as a structure-specific endonuclease that cleaves the replication fork D-loop intermediate, generating an accessible 3'-OH group in the leading-strand template for extension by DNA polymerase. Related references: PMID:22759634 PMID:28621305
  • In addition to endonuclease activity, ZRANB3 catalyzes fork regression via its annealing helicase activity to prevent replication fork disintegration and double-strand break formation. Related references: PMID:22704558 PMID:22705370
  • ZRANB3 localizes predominantly to the nucleus and is associated with chromosomes, consistent with its role in DNA replication and repair.
  • The protein undergoes alternative splicing and contains zinc-finger domains, which mediate DNA binding and interactions with ubiquitinated PCNA.
  • Post-translational modifications of ZRANB3 include phosphorylation and methylation, which may regulate its activity and stability.
  • ZRANB3 is classified under multiple functional categories, including ATP-binding, DNA damage, DNA repair, helicase, endonuclease, and metal-binding (zinc).

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the C-terminal region (amino acids 818-1077) of human ZRANB3, which may help avoid cross-reactivity with other zinc finger family members; consider this when interpreting results.
  • For Western blotting (WB), test the antibody in relevant cell or tissue lysates from human, mouse, or rat, as cross-reactivity has been noted for these species. Include proper loading and negative controls.
  • In ELISA applications, optimize coating antigen conditions and detection dilutions according to your assay requirements.
  • The predicted molecular weight is 123 kDa; confirm band migration with appropriate molecular weight markers and validate specificity using knockdown or knockout samples if available.
