Biological Background: ZP2 Function and Localization
- ZP2 (Zona pellucida sperm-binding protein 2), also known as zona pellucida glycoprotein 2 (Zp-2) and zona pellucida protein A, is an essential component of the oocyte extracellular matrix.
- As a structural component of the zona pellucida, ZP2 mediates sperm binding, triggers the acrosome reaction, and prevents post-fertilization polyspermy. It functions as a secondary sperm receptor. Related references: PMID:29895852
- The zona pellucida matrix is composed of 3 to 4 glycoproteins, including ZP1, ZP2, ZP3, and ZP4. Related references: PMID:29895852
- Subcellular localization: ZP2 is found in the zona pellucida surrounding the oocyte and is also associated with the cell membrane.
- Expression: ZP2 is specifically expressed in oocytes, as detected at the protein level.

Experimental Guidance and Technical Tips
- The immunogen (aa 651-745) represents the C-terminal region of human ZP2. When using this polyclonal antibody in Western blotting, a band around 82 kDa (predicted molecular weight) may be detected in lysates from human, mouse, or rat oocytes. However, cross-species reactivity should be validated individually.
- For immunofluorescence (IF-P), consider testing on ovarian tissue sections or isolated oocytes. Optimization of fixation and permeabilization conditions is recommended.
- ELISA applications may be useful for detecting ZP2 in solution; include appropriate controls to ensure specificity.
- As a polyclonal antibody, lot-to-lot consistency may vary. It is advisable to titrate the antibody for each new batch and application.
