Biological Background: ZO-1 Function and Localization
- ZO-1 (Tight junction protein 1) is a scaffolding protein belonging to the TJP family, which includes TJP2 and TJP3, that links tight junction transmembrane proteins (claudins, junctional adhesion molecules, occludin) to the actin cytoskeleton.
Related references: PMID:7798316 PMID:9792688 - The protein forms multistranded condensates that elongate into the tight junction belt, anchored apically via PATJ interaction, essential for epithelial polarization, barrier formation, and lumenogenesis.
- ZO-1 plays a regulatory role in cell migration by targeting CDC42BPB to the leading edge, and is important for podosome formation, regulating cell adhesion and matrix remodeling.

Experimental Guidance and Technical Tips
- The immunogen targets amino acids 18–273 of human ZO-1 (N-terminal region, spanning PDZ domains). Consider validating specificity with knockout or knockdown controls.
- For Western blot, expect a band at ~195 kDa. Use an appropriate gel percentage (e.g., 6–8%) and optimize transfer conditions for high-molecular-weight proteins.
- In immunofluorescence on paraffin-embedded sections (IF-P), optimize antigen retrieval; verify localization at tight junctions or membrane regions.
- This antibody cross-reacts with mouse and rat; suitable for rodent model studies; however, confirm signal in the relevant tissue before large-scale use.
