Biological Background: ZNF598 Function and Localization
- ZNF598 (also known as Zinc finger protein 598) is an E3 ubiquitin-protein ligase that serves as a ribosome collision sensor in the ribosome quality control (RQC) pathway, recognizing collided di-ribosomes to trigger translational arrest. Related references: PMID:28065601 PMID:28132843 PMID:28685749
- Upon stalled ribosome recognition, ZNF598 catalyzes monoubiquitination of 40S ribosomal proteins RPS10/eS10 and RPS3/uS3, as well as 'Lys-63'-linked polyubiquitination of RPS20/uS10, which facilitates recruitment of the RQT complex for ribosome disassembly. Related references: PMID:28065601 PMID:28132843 PMID:28685749 PMID:32099016 PMID:32579943 PMID:36302773
- The E3 ubiquitin ligase activity of ZNF598 is dependent on the E2 ubiquitin-conjugating enzyme UBE2D3, and the protein is specifically activated upon translation of prematurely polyadenylated mRNAs. Related references: PMID:28065601 PMID:28132843 PMID:28685749
- Independent of its RQC function, ZNF598 acts as an adapter that recruits the 4EHP-GYF2 complex to mRNAs and negatively regulates interferon-stimulated gene (ISG) expression. Related references: PMID:22751931 PMID:32726578 PMID:29719242
- During poxvirus infection, ZNF598 is required for viral protein synthesis by mediating ubiquitination of RPS10/eS10 and RPS20/uS10, likely enhancing translational efficiency on unusual 5' poly(A) leaders. Related references: PMID:29719242
- ZNF598 also acts as a positive regulator of HIV-1 Tat stability, reducing 'Lys-48'-linked ubiquitination and proteasomal degradation of Tat to promote viral replication and latent reactivation.
- Subcellularly, ZNF598 is localized to the cytoplasm and cytosol, and it features multiple zinc-finger domains, a phosphoprotein motif, and alternative splicing (UniProt Q86UK7).
Experimental Guidance and Technical Tips
- The immunogen used spans amino acids 120–420 of human ZNF598, covering central zinc-finger domains; this region is expected to generate antibodies recognizing both full-length and potential alternative splicing isoforms.
- For Western blot analysis, target bands near the expected molecular weight of ~99 kDa; additional bands may arise from post-translational modifications or isoform variants—validate specificity with appropriate controls.
- In ELISA applications, consider optimizing coating antigen and antibody dilution to account for the polyclonal nature; cross-reactivity may be minimal with other species due to the limited homology outside human.
- Given ZNF598’s role in ribosome stalling and viral replication, functional assays (e.g., puromycin labeling or viral infection models) can complement antibody-based detection but require careful biological validation.
