Biological Background: ZNF451 Function and Localization
- ZNF451 (Zinc finger protein 451) is also known as E3 SUMO-protein ligase ZNF451, coactivator for steroid receptors (COASTER), and ZATT. It belongs to the estrogen receptor (ER)-associated protein family that interacts with steroid receptors.
- Acts as an E3 SUMO-protein ligase with a strong preference for SUMO2 and SUMO3, facilitating UBE2I/UBC9-mediated sumoylation of target proteins. Related references: PMID:26524493, PMID:26524494
- Plays a critical role in protein SUMO2 modification under stress conditions such as DNA damage and proteasome inhibition (in vitro), and is required for MCM4 sumoylation.
- Preferentially transfers an additional SUMO2 chain onto the SUMO2 consensus site Lys-11; has no activity with SUMO1. Related references: PMID:26524493
- Negatively regulates transcriptional activation by the SMAD4 complex in response to TGF-beta signaling and inhibits EP300-mediated acetylation of histone H3 at Lys-9. Related references: PMID:24324267
- Participates in regulating the transcription of androgen receptor (AR) targets. Related references: PMID:18656483
- Localizes predominantly to the nucleus, including nucleoplasm and PML bodies, consistent with its roles in transcription regulation and sumoylation.
- Keywords highlight features such as zinc-finger domains, alternative splicing, phosphorylation, methylation, and its involvement in Ubl conjugation pathways.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a central region (aa 600-900) of human ZNF451, which is expected to recognize multiple isoforms due to its polyclonal nature.
- For Western blotting, a band around 121 kDa is anticipated. Consider using a gradient gel and appropriate molecular weight markers for high-molecular-weight protein transfer and detection.
- Species reactivity is confirmed for human, mouse, and rat. Cross-reactivity with other species should be verified experimentally if needed.
- In ELISA, optimize coating antigen concentration and antibody dilution using the immunogen peptide or full-length protein as a positive control.
- The antibody’s performance in immunoprecipitation or immunofluorescence has not been characterized; these applications may require further validation.
- Given ZNF451’s involvement in stress-induced sumoylation, it may be beneficial to include stress controls (e.g., DNA damage or proteasome inhibition) in functional studies.
