Biological Background: ZNF259 Function and Localization
- ZPR1 (ZNF259) is a zinc finger protein that acts as a signaling molecule, transmitting proliferative growth signals from the cytoplasm to the nucleus, thereby positively regulating cell cycle progression (PubMed:29851065). Related references: PMID:29851065
- It plays a critical role in the localization and accumulation of the survival motor neuron protein SMN1 in subnuclear bodies, including gems and Cajal bodies.
- ZPR1 induces neuron differentiation and stimulates axonal growth and growth cone formation in spinal cord motor neurons.
- It is involved in the splicing of cellular pre-mRNAs.
- ZPR1 may be involved in H2O2-induced neuronal cell death.
- Subcellularly, ZPR1 localizes to the nucleus, nucleolus, gems, Cajal bodies, and the perinuclear region of the cytoplasm, as well as in cell projections like axons and growth cones.
- It is expressed in fibroblasts and shows reduced expression in spinal muscular atrophy (SMA) patient fibroblasts.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 360-459, so consider epitope mapping if precise recognition is required.
- For Western blotting, the predicted protein weight is 51 kDa; use appropriate molecular weight markers and validate in the relevant sample system.
- In immunohistochemistry (paraffin-embedded), antigen retrieval conditions may need optimization for the specific tissue.
- As the antibody cross-reacts with mouse samples, it may be suitable for studies in mouse models; verify performance in your assay.
