Biological Background: ZMYND8 Function and Localization
- ZMYND8 (MYND-type zinc finger-containing chromatin reader), also designated RACK7/PRKCBP1, is a multidomain protein that integrates histone modification readout with transcriptional regulation and DNA damage repair.
- Acts as a dual histone modification reader, recognizing combinatorial marks such as H3.1K36me2-H4K16ac and H3K4me1-H3K14ac, enabling context-dependent recruitment of corepressor or coactivator complexes. Related references: PMID:26655721, PMID:27477906, PMID:31965980
- Recruits the NuRD chromatin remodeling complex to sites of DNA damage and specific gene promoters, facilitating transcriptional repression and homologous recombination repair. Related references: PMID:25593309, PMID:27732854, PMID:30134174
- Promotes productive transcription elongation by RNA polymerase II via interaction with the P-TEFb complex, and positively regulates ATRA-responsive and neuronal genes (e.g., MAPT). Related references: PMID:26655721, PMID:30134174, PMID:35916866, PMID:36064715
- Functions as a tumor suppressor in breast and prostate cancers by inhibiting cell invasion and metastasis via transcriptional modulation. Related references: PMID:27477906, PMID:31965980, PMID:33323928
- Localizes predominantly to the nucleus and chromatin, with additional presence in the cytoplasm.
- Exhibits highest expression in brain, lung, pancreas, and placenta, and is detected in neurons but absent in astrocytes; also expressed in cutaneous T-cell lymphomas (CTCL).
- Contains characteristic domains including a bromodomain, MYND zinc finger, and PHD finger, and is subject to post-translational modifications such as acetylation, phosphorylation, and ubiquitination (UniProt keywords).
Experimental Guidance and Technical Tips
- The immunogen comprises the C-terminal region (aa 959-1188), which may allow detection of full-length ZMYND8 and possibly various isoforms; verify in relevant cell or tissue lysates.
- For Western blot, the predicted molecular weight is ~132 kDa; use appropriate gel percentage and transfer conditions for high-molecular-weight proteins, and include KO-validation controls where available.
- In immunoprecipitation and ELISA, optimize antibody and lysate concentrations, and confirm specificity through reciprocal IP-western or competitive peptide blocking.
