Biological Background: ZMYND11 Function and Localization
- ZMYND11 (Zinc finger MYND domain-containing protein 11), also known as BS69 or BRAM1, is a chromatin reader that specifically recognizes histone H3.3 trimethylated at Lys-36 (H3.3K36me3), distinguishing it from canonical H3.1/H3.2.
- It functions as a transcription corepressor by modulating RNA polymerase II elongation at highly expressed genes.
- ZMYND11 binds non-specifically to double-stranded DNA, as demonstrated in vitro. Related references: PMID:24675531
- Acts as a tumor suppressor by repressing a transcriptional program essential for tumor cell growth.
- Inhibits Epstein-Barr virus EBNA2-mediated transcriptional activation and host cell proliferation during infection.
- Localizes predominantly to the nucleus and associates with chromosomes.
- Ubiquitously expressed across tissues.
- Contains a bromodomain and a MYND-type zinc finger domain; alternative splicing produces multiple isoforms.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 278–567 of human ZMYND11, spanning the MYND domain and adjacent regions; consider this when predicting epitope accessibility in different applications.
- Recommended for Western blot (WB) detection of endogenous ZMYND11 (predicted ~71 kDa) in human and mouse cell lysates; include appropriate controls and validate in your sample system.
- ELISA may be used to assess antibody titer or to screen for antigen binding; optimization of coating and detection conditions is advised.
- For immunofluorescence or other untested applications, pilot experiments are recommended to determine suitability.
