Biological Background: ZIP14 Function and Localization
- ZIP14, encoded by the SLC39A14 gene, is a ubiquitously expressed metal cation symporter belonging to the LIV-1 subfamily of ZIP zinc transporters (also known as LZT-Hs4). It mediates cellular uptake of zinc, manganese, and iron, which are vital for tissue homeostasis, metabolism, development, and immunity. - It functions as an electroneutral transporter, moving a divalent metal cation together with two bicarbonate anions across the plasma membrane. In addition to endogenous metals, ZIP14 can also import the toxic metal cadmium.
- ZIP14 plays a critical role in systemic manganese homeostasis by mediating basolateral manganese uptake in intestinal epithelial cells and uptake at the blood-brain barrier. Related references: PMID:31028174, PMID:31699897
- The protein is localized to the plasma membrane (apical and basolateral domains) as well as to early endosome, late endosome, and lysosome membranes, consistent with its roles in both cell-surface uptake and intracellular trafficking of metals.

- For Western blotting, a band around 54 kDa is predicted under reducing conditions; validation in the relevant sample system is recommended.
