- ZFYVE9 (Zinc finger FYVE domain-containing protein 9), also known as SARA (Smad anchor for receptor activation), is an early endosomal adaptor protein that belongs to the FYVE domain-containing family.
- It functions primarily in the TGF-β signaling pathway by recruiting SMAD2 and SMAD3 to the TGF-β receptor at the early endosome membrane, facilitating their phosphorylation and subsequent nuclear translocation.
- The protein modulates the transcriptional activity of the SMAD3/SMAD4 complex, thereby regulating downstream gene expression in response to TGF-β stimulation.
- ZFYVE9 may also be involved in TGF-β receptor internalization, contributing to signal attenuation and receptor trafficking.
- Subcellularly, ZFYVE9 is localized to the cytoplasm and specifically to the cytoplasmic side of the early endosome membrane, where its FYVE domain binds phosphatidylinositol 3-phosphate.
