Biological Background: ZFP36 Function and Localization
- ZFP36 (tristetraprolin, TTP) is a zinc-finger RNA-binding protein encoded by the ZFP36 gene (also known as G0S24, NUP475, TIS11). It belongs to the TIS11 family of tandem zinc-finger proteins.
- ZFP36 binds to AU-rich elements (AREs) in the 3' untranslated regions of target mRNAs, promoting their deadenylation and subsequent degradation via recruitment of the CCR4-NOT deadenylase complex and cytoplasmic decay machinery. This post-transcriptional regulation reduces protein synthesis of cytokines, growth factors, and other regulatory molecules. - ZFP36 binds its own mRNA and promotes its degradation, creating an autoregulatory feedback loop. Related references: PMID:15187101
- ZFP36 shuttles between the nucleus and cytoplasm and localizes to processing bodies (P-bodies) where it delivers target mRNAs for degradation.
