Biological Background: ZEB1 Function and Localization
- ZEB1 (Zinc finger E-box-binding homeobox 1), also known as TCF8, NIL-2-A, and DELTAEF1, is a transcription factor that contains zinc finger domains and a homeobox, enabling DNA binding.
- It functions primarily as a transcriptional repressor, inhibiting interleukin-2 (IL-2) expression and repressing E-cadherin promoter activity to promote epithelial-mesenchymal transition (EMT) via SMARCA4/BRG1 recruitment.
- ZEB1 can also act as a transcriptional activator, enhancing or repressing ATP1A1 promoter activity in a context-dependent manner, and positively regulates neuronal differentiation.
- In the absence of TGFB1, ZEB1 represses COL1A2 transcription by binding to E-box elements (5'-CANNTG-3') in the upstream enhancer.
- ZEB1 localizes to the nucleus, consistent with its role as a DNA-binding transcription factor.
- It is expressed in heart and skeletal muscle, and colocalizes with SMARCA4/BRG1 in E-cadherin-negative cells, such as those at the invasion front of colorectal carcinomas.
- Post-translational modifications include phosphorylation and isopeptide bond formation (via Ubl conjugation), which may regulate its activity and stability.
- ZEB1 mutations are associated with corneal dystrophies, including Fuchs endothelial corneal dystrophy (FECD6) and posterior polymorphous corneal dystrophy (PPCD3).
Experimental Guidance and Technical Tips
- This antibody was generated using a synthetic peptide immunogen spanning amino acids 800-900 of human ZEB1 (NP_110378.3). The polyclonal nature may recognize multiple epitopes, but cross-reactivity with other isoforms or related proteins should be validated in your system.
- For Western blotting, ZEB1 has a predicted molecular weight of 124 kDa. Use a protein ladder with appropriate high-molecular-weight markers and consider using a positive control such as nuclear extracts from cell lines expressing ZEB1 (e.g., some cancer lines).
- In immunofluorescence/ICC and IHC-P, nuclear localization is expected. Optimize permeabilization and blocking steps to enhance detection.
- ELISA applications may require optimization of coating antigen and detection conditions. The antibody is also reactive with mouse and rat, so it is suitable for cross-species studies.
