Biological Background: ZEB1 Function and Localization

  • ZEB1 is a zinc finger E-box-binding homeobox transcription factor that acts predominantly as a transcriptional repressor, regulating key developmental and disease processes.
  • It plays a critical role in epithelial-mesenchymal transition (EMT) by repressing the E-cadherin promoter through recruitment of SMARCA4/BRG1, promoting invasive phenotypes.
  • ZEB1 negatively regulates interleukin-2 (IL-2) gene expression and modulates the ATP1A1 promoter context-dependently, enhancing or repressing activity based on cellular conditions.
  • In the absence of TGFB1, it represses COL1A2 transcription via E-box binding; it also represses BCL6 transcription in the presence of the corepressor CTBP1.
  • ZEB1 positively regulates neuronal differentiation and represses RCOR1 transcription during neurogenesis, highlighting its role in neural development.
  • It localizes strictly to the nucleus and exhibits tissue-specific expression: detected in heart and skeletal muscle, absent from liver, spleen, and pancreas. At the protein level, it colocalizes with SMARCA4/BRG1 in E-cadherin-negative cells of colorectal carcinomas.
  • The protein contains homeobox and zinc-finger domains, undergoes phosphorylation, ubiquitination, and alternative splicing, and is associated with diseases such as corneal dystrophy.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to a synthetic peptide within amino acids 800–900 of human ZEB1; for Western blot, nuclear extracts are recommended given the target’s nuclear localization.
  • For ELISA applications, the peptide-based design may be suitable for direct detection; confirm performance with recombinant ZEB1 or cell lysates from expressing lines.
  • Cross-reactivity is reported for human and rat; validation in other species should be performed independently.
  • Optimize antibody dilution and blocking conditions for your specific assay system; consider using a known nuclear protein loading control for WB.
