Biological Background: ZDHHC5 Function and Localization

  • ZDHHC5 (Zinc finger DHHC domain-containing protein 5), also known as ZNF375, is a member of the DHHC palmitoyltransferase family.
  • ZDHHC5 is a palmitoyltransferase that adds palmitate to substrates including CTNND2, CD36, GSDMD, NLRP3, NOD1, NOD2, STAT3, and S1PR1, regulating cell adhesion, inflammation, fatty acid uptake, bacterial sensing, and cardiac functions. Related references: PMID:21820437 PMID:29185452 PMID:31402609
  • In synapses, ZDHHC5 palmitoylates delta-catenin/CTNND2 to enhance synaptic delivery and stabilization of AMPA receptors, with its membrane localization regulated by FYN phosphorylation and neuronal activity-dependent internalization. Related references: PMID:26334723
  • ZDHHC5 contributes to innate immunity by palmitoylating NOD1 and NOD2, facilitating their recruitment to bacterial entry sites and phagosomes. Related references: PMID:31649195 PMID:34293401
  • In fatty acid uptake, ZDHHC5 palmitoylates CD36, targeting it to the plasma membrane; subsequent fatty acid binding triggers LYN-mediated phosphorylation and inactivation of ZDHHC5, leading to CD36 endocytosis. Related references: PMID:32958780
  • ZDHHC5 regulates inflammatory responses by palmitoylating NLRP3 to promote inflammasome assembly and activation, and by palmitoylating gasdermin-D (GSDMD) to promote its membrane translocation and pyroptotic pore formation. Related references: PMID:38092000 PMID:38530158 PMID:38599239
  • ZDHHC5 controls oligodendrocyte development via STAT3 palmitoylation and may contribute to sperm tail assembly by palmitoylating IFT81 during spermatogenesis.
  • ZDHHC5 localizes to the cell membrane, synapse, and Golgi apparatus. It is a multi-pass transmembrane protein subject to alternative splicing, methylation, and phosphorylation.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 130-230 of human ZDHHC5, a region that may be conserved in mouse; the stated cross-reactivity with mouse should be validated in appropriate sample systems.
  • For Western blotting, consider the predicted molecular weight of approximately 78 kDa. Validate band specificity using appropriate controls and, if necessary, peptide blocking experiments.
  • For ELISA applications, the immunizing peptide can serve as a useful positive control to assess antibody binding.
