Biological Background: ZDHHC2 Function and Localization

  • ZDHHC2, also known as DHHC2, ZNF372, REC and REAM, is a member of the DHHC family of palmitoyltransferases that catalyze the attachment of palmitate to cysteine residues on target proteins.
  • It exhibits broad fatty acid selectivity, transferring not only palmitate but also myristate and stearate onto substrates, thereby regulating protein trafficking, membrane localization and function.
    Related references: PMID:18296695 / PMID:18508921 / PMID:19144824
  • In the nervous system, ZDHHC2 palmitoylates AKAP5, DLG4 and RGS7BP, playing a key role in synaptic plasticity by controlling AMPA receptor clustering and GPCR signaling in dendrites.
  • ZDHHC2 regulates cell adhesion by palmitoylating CD9 and CD151, thereby controlling their surface expression and function.
    Related references: PMID:18508921
  • It palmitoylates the endoplasmic reticulum protein CKAP4, promoting its translocation to the plasma membrane, and may also modify LCK to influence immune signaling.
    Related references: PMID:18296695 / PMID:19144824 / PMID:22034844
  • Subcellularly, ZDHHC2 localizes to the postsynaptic density, recycling endosomes, cell membrane, ER and Golgi membranes, consistent with its roles in protein trafficking and signaling.
  • ZDHHC2 is ubiquitously expressed, with notably reduced levels observed in colorectal cancers with liver metastasis, linking it to tumor progression.
    Related references: PMID:10918388 / PMID:22034844

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 270–367 of human ZDHHC2. The resulting antiserum is expected to recognize full-length ZDHHC2 and isoforms containing this region; however, cross-reactivity with other family members should be assessed.
  • For Western blotting, a band of approximately 42 kDa is anticipated. Include appropriate controls (e.g., ZDHHC2‑overexpressing lysates) and optimize sample preparation for this multi‑pass transmembrane protein.
  • In ELISA, the antibody can serve as a capture or detection reagent. Pair with a compatible secondary antibody and validate with recombinant protein or known positive samples.
  • Since ZDHHC2 is palmitoylated and membrane‑associated, consider using detergent‑containing lysis buffers and avoid boiling if epitope sensitivity is a concern.
