Biological Background: Palmitoyltransferase ZDHHC15 Function and Localization
- ZDHHC15 is a member of the zinc finger DHHC domain-containing protein family, functioning as an acyltransferase that catalyzes protein palmitoylation.
- It mediates the addition of palmitate onto various protein substrates and can also transfer myristate and stearate, displaying broad fatty acid selectivity.
- Palmitoylates IGF2R and SORT1, facilitating their partitioning to endosomal membrane subdomains where they interact with the retromer cargo-selective complex, thereby regulating retrograde transport of lysosomal sorting receptors from endosomes to the Golgi. Related references: PMID:18817523
- In the nervous system, it palmitoylates DLG4/PSD95 to regulate synaptic clustering and synaptogenesis, and may also palmitoylate GAP43, DNAJC5, CALHM3, and FYN (PubMed:19956733); involved in dopaminergic neuron differentiation and diencephalon development. Related references: PMID:19956733
- Localizes predominantly to the Golgi apparatus membrane and postsynaptic density.
