Biological Background: ZC3H12D Function and Localization
- ZC3H12D (also known as MCPIP4, p34, TFL) is a probable ribonuclease featuring a CCCH-type zinc finger domain, and it belongs to the ZC3H12 family of RNases.
- It may regulate cell growth by suppressing RB1 phosphorylation, thereby inhibiting G1 to S phase progression; this activity suggests a tumor suppressor role. Related references: PMID:19531561
- It serves as a tumor suppressor in certain leukemia cells. Related references: PMID:17210687
- Together with ZC3H12A, it enhances the degradation of interleukin IL-6 mRNA in activated macrophages, indicating a role in inflammatory response regulation. Related references: PMID:26134560
- ZC3H12D localizes predominantly to the cytoplasm and P-bodies, and also shuttles to the nucleus, consistent with its role in mRNA decay.
- Expression is found in normal lymphocytes but is defective in some leukemia/lymphoma cell lines, implicating it in lymphocyte biology and hematopoietic malignancies.
- It binds metal ions, likely magnesium, which is characteristic of its nuclease activity.
- Alternative splicing and chromosomal rearrangement events are associated with this gene, possibly contributing to its dysregulation in cancer.
Experimental Guidance and Technical Tips
- For WB, this polyclonal antibody may detect endogenous ZC3H12D at ~58 kDa. Consider using whole-cell extracts from lymphocyte or leukemia cell lines as positive controls.
- For IF/ICC, expect cytoplasmic and nuclear staining; P-body localization may be observed under specific conditions. Validate fixation/permeabilization methods for optimal results.
- For ELISA, the antibody can be paired with appropriate detection reagents; consider titration for best signal-to-noise ratio.
- Since the immunogen corresponds to a recombinant fusion protein within the central region (aa150-230), the antibody may recognize multiple isoforms containing this epitope; confirm specificity by siRNA knockdown or using knockout models if available.
- Cross-reactivity with mouse and rat is reported; validate in the species-specific experimental context.
