Biological Background: Endoribonuclease ZC3H12A Function and Localization
- Endoribonuclease ZC3H12A, also known as MCPIP-1, Regnase-1, or Reg1, is a zinc-finger CCCH domain-containing protein with dual endoribonuclease and deubiquitinase activities.
- It modulates inflammatory responses by promoting degradation of translationally active cytokine-induced mRNAs such as IL6 and IL12B during early inflammation (PubMed:26320658), and cleaves mRNAs via stem-loop structures in a UPF1-dependent manner (PubMed:19909337, PubMed:22561375, PubMed:26134560). - ZC3H12A prevents aberrant T-cell activation by degrading mRNAs encoding cytokines, cell surface receptors, and transcription factors, and cooperatively inhibits Th17 cell differentiation with RC3H1 (By similarity).
- It inhibits microRNA biogenesis by cleaving terminal loops of precursor miRNAs important for inflammation, thereby preventing mature miRNA synthesis (PubMed:22055188). Related references: PMID:22055188
