Biological Background: ZBTB48 Function and Localization
- ZBTB48, also known as HKR3, TZAP, or ZNF855, is a zinc finger and BTB domain-containing protein that functions as a transcription regulator and telomere-binding protein.
- It plays a critical role in telomere length homeostasis by directly binding the double-stranded telomeric repeat 5'-TTAGGG-3', preferentially at telomeres with low shelterin complex occupancy, and initiating telomere trimming to prevent aberrantly long telomeres. Related references: PMID:28082411, PMID:28500257
- As a transcriptional regulator, ZBTB48 binds promoter regions and controls expression of genes such as MTFP1, the tumor suppressor ARF (CDKN2A isoform), and CIITA, a master regulator of MHC class II genes. Related references: PMID:24382891, PMID:28500257, PMID:7969177, PMID:39562739
- ZBTB48 also regulates cellular RNA m6A/m6Am methylation by facilitating recruitment of the RNA demethylase FTO to target mRNAs. Related references: PMID:39300486
- The protein localizes to the nucleus and specifically associates with telomeric chromatin.
- It is expressed in tissues such as adrenal gland and neuroblastoma, suggesting roles in these contexts.
- ZBTB48 contains multiple zinc finger domains and a BTB/POZ domain, characteristic of transcription factors, and is subject to post-translational modifications including phosphorylation and ubiquitination.
Experimental Guidance and Technical Tips
- The immunogen used to generate this antibody corresponds to the N-terminal 1–280 amino acids of human ZBTB48 (NP_005332.1), a region that includes the BTB domain and part of the zinc finger domain; consider verifying reactivity with full-length protein.
- For WB applications, test on lysates from human or mouse cells, especially those with reported ZBTB48 expression (e.g., adrenal gland, neuroblastoma).
- ELISA is also indicated; optimize coating antigen and detection conditions for your specific assay format.
- Since cross-reactivity is indicated for human and mouse, sequence alignment may help predict reactivity with other species.
