Biological Background: ZBTB17 Function and Localization

  • Also known as MIZ-1, ZNF151, and ZNF60, ZBTB17 is a transcription factor that can function as either an activator or repressor depending on its binding partners and the cellular context.
  • Plays a critical role in early lymphocyte development by preventing apoptosis in lymphoid precursors, enabling them to survive in response to IL7 and undergo proper lineage commitment.
  • Binds to the promoters of adenovirus major late protein and cyclin D1, thereby activating transcription.
  • Represses RB1 transcription; this repression can be blocked by interaction with ZBTB49 isoform 3. Related references: PMID:25245946
  • Required for early embryonic development during gastrulation.
  • Localizes to the nucleus where it exerts its transcriptional functions.
  • Expressed in germinal center B-cells, consistent with its role in lymphocyte biology.
  • Contains BTB and zinc finger domains; subject to phosphorylation and ubiquitination, classified as a DNA-binding, metal-binding phosphoprotein with isopeptide bonds.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 121–360 of human ZBTB17, a region likely to be accessible for antibody binding. Consider using the recombinant fusion protein as a positive control in assays.
  • Predicted molecular weight is 88 kDa; for Western blot, validate the specific band size in your sample system and consider loading appropriate positive and negative controls.
  • Cross-reactivity with mouse and rat indicates conserved epitopes; however, validation in the specific tissue or cell line is recommended before interpreting results.
