Biological Background: ZBP1 Function and Localization

  • ZBP1 (Z-DNA-binding protein 1), also known as DAI or DLM-1, is a key innate immune sensor that recognizes Z-RNA structures produced during viral infections, triggering programmed cell death pathways including necroptosis, apoptosis, and pyroptosis (PANoptosis). Related references: PMID:32200799
  • Upon Z-RNA binding, ZBP1 interacts with RIPK3 kinase to initiate MLKL phosphorylation and necroptosis, and also activates caspase-8 and the NLRP3 inflammasome for apoptosis and pyroptosis, respectively.
