Biological Background: ZBP1 Function and Localization
- ZBP1 (Z-DNA-binding protein 1), also known as DAI (DNA-dependent activator of IFN-regulatory factors) and DLM-1, is a 46kDa innate immune sensor that specifically recognizes Z-conformation nucleic acids, particularly Z-RNA structures produced during viral infection.
- Functions as a key mediator of PANoptosis (pyroptosis, apoptosis, necroptosis), an integrated cell death pathway defending against pathogens. It activates RIPK3, caspase-8, and the NLRP3 inflammasome to orchestrate inflammatory cell death. Related references: PMID:32200799
- Acts as a critical activator of necroptosis: upon binding Z-RNA, ZBP1 interacts with and stimulates RIPK3, which phosphorylates MLKL, triggering execution of programmed necrosis.
