Biological Background: ZBP1 Function and Localization

  • ZBP1 (Z-DNA-binding protein 1), also known as DAI (DNA-dependent activator of IFN-regulatory factors) and DLM-1, is a 46kDa innate immune sensor that specifically recognizes Z-conformation nucleic acids, particularly Z-RNA structures produced during viral infection.
  • Functions as a key mediator of PANoptosis (pyroptosis, apoptosis, necroptosis), an integrated cell death pathway defending against pathogens. It activates RIPK3, caspase-8, and the NLRP3 inflammasome to orchestrate inflammatory cell death. Related references: PMID:32200799
  • Acts as a critical activator of necroptosis: upon binding Z-RNA, ZBP1 interacts with and stimulates RIPK3, which phosphorylates MLKL, triggering execution of programmed necrosis.
  • During influenza A virus (IAV) infection, ZBP1 drives nuclear necroptosis by sensing Z-RNA in the nucleus, leading to RIPK3-MLKL activation, nuclear envelope disruption, and leakage of cellular DNA into the cytosol. Related references: PMID:32200799
  • Component of the AIM2 PANoptosome complex that triggers PANoptosis in response to bacterial infection; also serves as the apical sensor of fungal infection activating PANoptosis.
  • In some cell types, ZBP1 restricts viral replication via cell death-independent mechanisms. For example, in Zika virus-infected neurons, ZBP1-RIPK3 upregulate the enzyme ACOD1/IRG1, producing the metabolite itaconate which suppresses viral genome replication.
  • Subcellular localization includes both cytoplasm and nucleus, consistent with its roles in sensing cytosolic viral RNA and nuclear Z-RNA during orthomyxovirus infection.
  • Highly expressed in lymphatic tissues (lymph node, leukocytes, tonsil, bone marrow, spleen); also detected in thymus, lung, and liver. Related references: PMID:11842111

Experimental Guidance and Technical Tips

  • The immunogen covers the N-terminal region (amino acids 1–170) containing the Zα and Zβ DNA-binding domains, likely enabling recognition of full-length ZBP1 and possibly some splice variants. Verify reactivity with the relevant sample system.
  • For western blotting (WB), the predicted molecular weight is approximately 46 kDa; users should consider using positive controls such as virus-infected or stimulated immune cell lines (e.g., THP-1, U937) to confirm specificity.
  • In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), fixation and permeabilization conditions should be optimized, as ZBP1 can localize to both cytoplasm and nucleus depending on activation status.
  • For immunoprecipitation (IP) and ELISA applications, validate antibody performance under non-denaturing conditions and consider crosslinking to solid supports if needed.
  • Since this antibody is validated for human species, cross-reactivity with mouse or rat ZBP1 is not guaranteed; users requiring reactivity in other species should assess independently.
