Biological Background: ZBP1 Function and Localization
- ZBP1, also known as DAI or DLM-1, is a Z-DNA/RNA-binding protein that acts as a key innate immune sensor of viral infections. It recognizes Z-RNA structures produced by viruses such as herpesvirus, orthomyxovirus, and flavivirus, triggering PANoptosis (pyroptosis, necroptosis, and apoptosis) to eliminate infected cells. - It mediates necroptosis by binding Z-RNA and then interacting with RIPK3, which phosphorylates MLKL, leading to membrane rupture and cell death. This pathway is critical in antiviral defense, particularly against orthomyxoviruses like influenza A virus (IAV) where nuclear Z-RNA forms trigger necroptosis. - For Western blotting, consider the predicted molecular weight of 44 kDa and be aware that post-translational modifications (e.g., ubiquitination) may shift the band; validate with appropriate positive and negative controls.
- In immunohistochemistry and immunofluorescence, optimize fixation/permeabilization for nuclear and cytoplasmic detection; use isotype controls and confirm expression in mouse tissues such as lung, spleen, or activated macrophage cultures.
- For ELISA, the polyclonal nature may offer high sensitivity; we recommend pairing with a capture antibody and validating against known positive samples to establish assay parameters.
