Biological Background: ZBP1 Function and Localization

  • ZBP1, also known as DAI or DLM-1, is a Z-DNA/RNA-binding protein that acts as a key innate immune sensor of viral infections. It recognizes Z-RNA structures produced by viruses such as herpesvirus, orthomyxovirus, and flavivirus, triggering PANoptosis (pyroptosis, necroptosis, and apoptosis) to eliminate infected cells. Related references: PMID:17618271, PMID:18375758, PMID:19590578, PMID:27746097, PMID:27917412, PMID:28607035
  • It mediates necroptosis by binding Z-RNA and then interacting with RIPK3, which phosphorylates MLKL, leading to membrane rupture and cell death. This pathway is critical in antiviral defense, particularly against orthomyxoviruses like influenza A virus (IAV) where nuclear Z-RNA forms trigger necroptosis. Related references: PMID:22423968, PMID:27819681, PMID:32200799, PMID:32296175
  • ZBP1-dependent cell death in IAV infection also promotes IL1A induction, which synergizes with IL1B to recruit neutrophils and form neutrophil extracellular traps, aiding in pathogen clearance. Related references: PMID:31630209
  • Beyond necroptosis, ZBP1 is a component of the AIM2 PANoptosome complex that activates PANoptosis in response to bacterial and fungal infections. Related references: PMID:34471287, PMID:33109609
  • ZBP1 can also trigger CASP8-mediated cell death independently of Z-RNA sensing, through interaction with RIPK1. Related references: PMID:33397971
  • In certain contexts, ZBP1 restricts viral replication via cell death-independent mechanisms: in neurons infected with flavivirus, ZBP1 and RIPK3 upregulate ACOD1/IRG1, producing itaconate that inhibits succinate dehydrogenase and suppresses viral genome replication. Related references: PMID:30635240
  • ZBP1 is localized in both the cytoplasm and nucleus, enabling it to detect Z-RNA in different cellular compartments.
  • It is predominantly expressed in lung, spleen, and liver, with lower levels in heart, kidney, and testis; expression is upregulated in tumor stromal cells and activated macrophages.

Experimental Guidance and Technical Tips

  • The immunogen covers the N-terminal region (aa 1–160), which contains Z-DNA-binding domains; this antibody may detect full-length (44 kDa) and N-terminal isoforms, as well as an alternative splice form migrating at ~21 kDa.
  • For Western blotting, consider the predicted molecular weight of 44 kDa and be aware that post-translational modifications (e.g., ubiquitination) may shift the band; validate with appropriate positive and negative controls.
  • In immunohistochemistry and immunofluorescence, optimize fixation/permeabilization for nuclear and cytoplasmic detection; use isotype controls and confirm expression in mouse tissues such as lung, spleen, or activated macrophage cultures.
  • For ELISA, the polyclonal nature may offer high sensitivity; we recommend pairing with a capture antibody and validating against known positive samples to establish assay parameters.
