Biological Background: ZBED1 Function and Localization
- ZBED1 (also known as DREF, hDREF, TRAMP, ALTE) is an E3 SUMO ligase that contains a BED-type zinc finger domain and belongs to the ZBED family of DNA-binding proteins.
- It catalyzes the SUMOylation of CHD3/Mi2-alpha, causing its release from DNA and alleviating transcriptional repression. Related references: PMID:27068747
- ZBED1 binds the consensus sequence 5'-TGTCG[CT]GA[CT]A-3' in gene promoters, including those of ribosomal proteins (RPS6, RPL10A, RPL12) and histone H1-5, thereby enhancing their transcription. Related references: PMID:12663651, PMID:17209048, PMID:17220279
- This transcriptional activation promotes cell proliferation. Related references: PMID:12663651, PMID:17220279
- The protein localizes to the nucleus and PML nuclear bodies.
- ZBED1 is ubiquitously expressed at low levels, with highest expression in skeletal muscle, heart, spleen, and placenta.
- During adenovirus infection, ZBED1 binds to viral promoters and contributes to transcriptional repression, inhibiting early-stage virus growth.
- Structurally, ZBED1 features a BED-type zinc finger DNA-binding domain and functions as a SUMO transferase, playing key roles in transcription regulation and host-virus interactions.
Experimental Guidance and Technical Tips
- The immunogen spans the N-terminal region (amino acids 1–280) of human ZBED1; reactivity with all isoforms should be validated if multiple variants are expected.
- For Western blotting, a band at approximately 78 kDa is anticipated; use appropriate molecular weight markers and a positive control lysate (e.g., HEK293 or HeLa cells).
- When performing IHC-P or IF/ICC, optimize fixation and antigen retrieval conditions; nuclear localization may require permeabilization steps.
- ELISA assays can be validated using the recombinant immunogen protein as a positive control.
- This antibody is reported to react with human ZBED1; for other species, perform sequence alignment and pilot testing prior to use.
- Consider subcellular fractionation to enrich nuclear proteins for enhanced detection in WB or IF.
