- Tyrosine-protein kinase ZAP-70 (also known as SRK, Syk-related tyrosine kinase) plays an essential role in adaptive immune response regulation.
- Regulates T-cell motility, adhesion, cytokine expression, and thymocyte development; also contributes to primary B-lymphocyte development and activation.
- Recruited to phosphorylated T-cell receptor (TCR) components CD3E and CD247/CD3Z via ITAM motif at the plasma membrane, leading to relief of autoinhibition (PubMed:7509083). Related references: PMID:7509083
- Phosphorylates key adapter proteins LAT and LCP2, initiating a signaling cascade that promotes T-cell proliferation, differentiation, and lymphokine production.
- Controls cytoskeleton modifications, adhesion, and mobility of T-lymphocytes to ensure correct effector delivery to antigen-presenting cells.
- Required for TCR-CD247/CD3Z internalization and degradation through interaction with E3 ubiquitin-protein ligase CBL and adapters SLA and SLA2, modulating TCR surface expression.
