Biological Background: ZAP70 Function and Localization

  • Tyrosine-protein kinase ZAP-70 (also known as SRK, Syk-related tyrosine kinase) plays an essential role in adaptive immune response regulation.
  • Regulates T-cell motility, adhesion, cytokine expression, and thymocyte development; also contributes to primary B-lymphocyte development and activation.
  • Recruited to phosphorylated T-cell receptor (TCR) components CD3E and CD247/CD3Z via ITAM motif at the plasma membrane, leading to relief of autoinhibition (PubMed:7509083). Related references: PMID:7509083
  • Phosphorylates key adapter proteins LAT and LCP2, initiating a signaling cascade that promotes T-cell proliferation, differentiation, and lymphokine production.
  • Controls cytoskeleton modifications, adhesion, and mobility of T-lymphocytes to ensure correct effector delivery to antigen-presenting cells.
  • Required for TCR-CD247/CD3Z internalization and degradation through interaction with E3 ubiquitin-protein ligase CBL and adapters SLA and SLA2, modulating TCR surface expression.
  • Localizes to the cytoplasm and cell membrane.
  • Primarily expressed in T-cells, natural killer cells, early thymocytes, and pro/pre B-cells.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 520–619 near the C-terminus; consider that epitope may be conserved in isoforms or truncated forms.
  • For Western blot, expect a band approximately 70 kDa; validate using T-cell lines (e.g., Jurkat) or mouse lymphoid tissues as positive controls.
  • In ELISA, verify specificity against recombinant ZAP70 and avoid cross-reactivity with related Syk kinase; optimize coating and detection conditions.
  • Post-translational modifications (phosphorylation, ubiquitination) may affect antibody binding; perform appropriate controls in your experimental system.
