Biological Background: ZAP70 Function and Localization
- ZAP70 (Tyrosine-protein kinase ZAP-70) is a 70 kDa zeta-chain associated protein, also known as Syk-related tyrosine kinase, encoded by the ZAP70 (SRK) gene.
- Plays an essential role in the adaptive immune response by regulating T‑cell motility, adhesion, cytokine expression, and thymocyte development. Upon TCR activation, ZAP70 is recruited to phosphorylated CD3 chains via ITAM motifs at the plasma membrane. Related references: PMID:7509083
- Subcellular localization is primarily cytoplasmic, with translocation to the cell membrane following TCR engagement.
- Tissue expression is restricted to T‑cells, natural killer cells, early thymocytes, and pro/pre B‑cells.
