Biological Background: ZAP70 Function and Localization

  • ZAP70 (Tyrosine-protein kinase ZAP-70) is a 70 kDa zeta-chain associated protein, also known as Syk-related tyrosine kinase, encoded by the ZAP70 (SRK) gene.
  • Plays an essential role in the adaptive immune response by regulating T‑cell motility, adhesion, cytokine expression, and thymocyte development. Upon TCR activation, ZAP70 is recruited to phosphorylated CD3 chains via ITAM motifs at the plasma membrane. Related references: PMID:7509083
  • Subcellular localization is primarily cytoplasmic, with translocation to the cell membrane following TCR engagement.
  • Tissue expression is restricted to T‑cells, natural killer cells, early thymocytes, and pro/pre B‑cells.
  • Contains N‑terminal tandem SH2 domains and a C‑terminal kinase domain; undergoes phosphorylation on multiple tyrosine residues and acetylation.
  • Also required for TCR internalization and degradation through interaction with E3 ubiquitin ligases, thereby modulating T‑cell activation switch-off.
  • Contributes to primary B‑lymphocyte development and activation through B‑cell receptor signaling.

Experimental Guidance and Technical Tips

  • The immunogen spans amino acids 1‑300 of human ZAP70 (N‑terminal region), covering the SH2 domains; reactivity against shorter isoforms or splice variants should be verified experimentally.
  • For Western blot, consider using whole‑cell lysates from Jurkat or human PBMCs; mouse or rat splenocyte lysates may also be suitable given the reported cross‑reactivity.
  • In immunofluorescence/ICC, validate fixation and permeabilization conditions in the relevant sample system, as the protein can be cytosolic or membrane‑associated.
  • ELISA applications may require optimization of coating conditions and antibody dilution; inclusion of a blocking step with recombinant ZAP70 protein could help assess specificity.
