Biological Background: YTHDF1 Function and Localization
- YTHDF1, also known as Dermatomyositis associated with cancer putative autoantigen 1 (DACA-1), is a YTH domain-containing protein that specifically recognizes and binds N6-methyladenosine (m6A)-modified mRNAs, regulating their stability and processing. - It promotes degradation of m6A-containing mRNAs by recruiting the CCR4-NOT complex, and functions redundantly with paralogs YTHDF2 and YTHDF3 to mediate mRNA decay and cellular differentiation. Related references: PMID:28106072, PMID:32492408
- YTHDF1 is involved in learning and memory formation in the hippocampus by binding to m6A-containing neuronal mRNAs, and regulates axon guidance via ROBO3 mRNA (by similarity).
- It acts as a negative regulator of antigen cross-presentation in dendritic cells, and in tumors this limits anti-tumor immune responses (by similarity).
