Biological Background: YTHDF1 Function and Localization

  • YTHDF1, also known as Dermatomyositis associated with cancer putative autoantigen 1 (DACA-1), is a YTH domain-containing protein that specifically recognizes and binds N6-methyladenosine (m6A)-modified mRNAs, regulating their stability and processing. Related references: PMID:24284625, PMID:26318451, PMID:32492408
  • It promotes degradation of m6A-containing mRNAs by recruiting the CCR4-NOT complex, and functions redundantly with paralogs YTHDF2 and YTHDF3 to mediate mRNA decay and cellular differentiation. Related references: PMID:28106072, PMID:32492408
  • YTHDF1 is involved in learning and memory formation in the hippocampus by binding to m6A-containing neuronal mRNAs, and regulates axon guidance via ROBO3 mRNA (by similarity).
  • It acts as a negative regulator of antigen cross-presentation in dendritic cells, and in tumors this limits anti-tumor immune responses (by similarity).
  • YTHDF1 undergoes liquid-liquid phase separation upon binding multivalent m6A-modified mRNAs, promoting partitioning into cytoplasmic membraneless compartments such as P-bodies and stress granules. Related references: PMID:31292544, PMID:31388144, PMID:32451507
  • Subcellular localization includes cytoplasm, P-bodies, and stress granules.
  • The protein is subject to acetylation, phosphorylation, and ubiquitination, and is identified as a phosphoprotein and Ubl conjugation target.

Experimental Guidance and Technical Tips

  • For Western blotting, the predicted molecular weight of full-length YTHDF1 is approximately 61 kDa; a smaller isoform may be observed at ~20 kDa. Use appropriate positive and negative controls to confirm specificity.
  • This antibody is suitable for immunoprecipitation and ELISA based on the manufacturer's data; optimize antibody concentration and incubation conditions for your specific sample type.
  • Cross-reactivity with mouse and rat makes it suitable for cross-species studies; however, validate performance in each species’ tissues/cells.
