Biological Background: YTHDF1 Function and Localization

  • YTHDF1 (YTH domain-containing family protein 1), also known as Dermatomyositis associated with cancer putative autoantigen 1 (DACA-1), is a member of the YTHDF paralog family (YTHDF1, YTHDF2, YTHDF3) that specifically binds N6-methyladenosine (m6A)-modified mRNAs.
  • Recognizes and binds m6A-containing mRNAs to regulate their stability and translation (PubMed:24284625, PubMed:26318451, PubMed:32492408). Related references: PMID:24284625
    PMID:26318451
    PMID:32492408
  • The YTHDF proteins act redundantly to mediate m6A-dependent mRNA degradation and cellular differentiation via the CCR4-NOT complex (PubMed:28106072, PubMed:32492408). Related references: PMID:28106072
    PMID:32492408
  • Localizes to the cytoplasm, where it can partition into membraneless compartments such as P-bodies and stress granules through liquid-liquid phase separation upon binding polymethylated mRNAs (PubMed:31292544, PubMed:31388144, PubMed:32451507). Related references: PMID:31292544
    PMID:31388144
    PMID:32451507
  • Plays a role in learning and memory formation in the hippocampus by binding m6A-containing neuronal mRNAs (By similarity).
  • Acts as a regulator of axon guidance via ROBO3 transcript binding (By similarity) and negatively regulates antigen cross-presentation in dendritic cells, which may limit anti-tumor immune responses (By similarity).
  • The gene YTHDF1 undergoes alternative splicing, producing isoforms with distinct functional properties.
  • Post-translationally modified by acetylation, phosphorylation, and ubiquitination (keywords: Acetylation, Phosphoprotein, Ubl conjugation).

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 280-390 within the C-terminal region of human YTHDF1, which includes part of the YTH domain; consider using it for Western blot (WB) detection of endogenous YTHDF1 (predicted ~61 kDa) across human, mouse, and rat samples.
  • For immunohistochemistry (IHC-P), validate antibody specificity in formalin-fixed paraffin-embedded tissues with appropriate controls, as YTHDF1 cytoplasmic localization may require antigen retrieval optimization.
  • In ELISA applications, the polyclonal nature may offer broad epitope recognition, but ensure cross-reactivity evaluation with other YTHDF family members if needed.
