Biological Background: YTHDF1 Function and Localization

  • YTHDF1 is a cytoplasmic m6A reader protein, also known as DACA-1, encoded by the YTHDF1 gene (C20orf21).
  • It specifically binds N6-methyladenosine (m6A)-modified mRNAs and regulates their stability by promoting degradation via the CCR4-NOT complex. Related references: PMID:24284625, PMID:26318451, PMID:32492408
  • YTHDF1 acts redundantly with paralogs YTHDF2 and YTHDF3 to mediate m6A-dependent mRNA degradation and cellular differentiation. Related references: PMID:28106072, PMID:32492408
  • It localizes to the cytoplasm, P-bodies, and stress granules; m6A binding triggers liquid-liquid phase separation, driving formation of membraneless compartments. Related references: PMID:31292544, PMID:31388144, PMID:32451507
  • Tissue-specific functions include facilitating hippocampal learning and memory, regulating axon guidance, and acting as a negative regulator of antigen cross-presentation in dendritic cells.
  • The protein is subject to acetylation, phosphorylation, and ubiquitination, as indicated by UniProt keywords.

Experimental Guidance and Technical Tips

  • The immunogen covers the N-terminal region (aa 1-200); verify specificity in your model system as reactivity may depend on protein conformation.
  • For WB, the predicted molecular weight is ~61 kDa; use appropriate gel percentage and control lysates from human, mouse, or rat cells.
  • For IF/ICC, evaluate fixation/permeabilization conditions to detect both diffuse cytoplasmic and granular localization.
  • For IP, include RNase treatment if RNA-dependent interactions are a concern.
  • Cross-reactivity with paralogs (YTHDF2, YTHDF3) may occur due to sequence similarity; consider isoform-specific validation.
  • Always include positive and negative controls for application-specific optimization.
