Biological Background: Ythdc1 Function and Localization
- Ythdc1 (YTH domain-containing protein 1) is a nuclear reader of N6-methyladenosine (m6A), a prevalent internal mRNA modification, and specifically recognizes and binds m6A-containing RNAs. Related references: PMID:29262316, PMID:29799838
- It acts as a key regulator of alternative splicing; it promotes exon inclusion by recruiting splicing factor SRSF3 to mRNA sites adjacent to m6A, while inhibiting exon skipping via competition with SRSF10.
- Ythdc1 facilitates nuclear export of m6A-modified mRNAs through interaction with SRSF3 and the export factor NXF1.
- It contributes to S-adenosyl-L-methionine homeostasis by regulating the expression of MAT2A, likely through binding m6A-methylated MAT2A transcripts. Related references: PMID:29262316
- Ythdc1 is essential for germline development: it is required for spermatogonial development in males and oocyte growth and maturation in females, at least in part through its role in alternative splicing. Related references: PMID:29799838
- Independently of its m6A reader activity, Ythdc1 protects the transcription factor PPARG from ubiquitin-mediated proteasomal degradation by preventing its interaction with the E3 ligase ARIH2, thereby supporting brown adipose tissue development and energy homeostasis. Related references: PMID:40355558
- Ythdc1 is localized predominantly in the nucleus and concentrates in nuclear speckles, consistent with its functions in mRNA processing and splicing.
- Additional reported functions include binding m6A on Xist RNA to promote X chromosome inactivation and recognizing m6A on single-stranded DNA; the protein is also subject to phosphorylation and ubiquitin-like modifications.
Experimental Guidance and Technical Tips
- This antibody targets the C-terminal region (aa 597–736) of mouse Ythdc1, which is highly conserved in human YTHDC1; cross-reactivity with human samples is expected.
- For Western blotting, use standard SDS-PAGE and PVDF membrane transfer under reducing conditions. A band near 85 kDa should be observed; post-translational modifications may cause slight shifts.
- As a KO-validated antibody, it is recommended to include appropriate knockout cell or tissue lysates as negative controls to confirm specificity.
- For ELISA applications, consider using the recombinant protein immunogen for assay setup and optimization.
- Given the sequence conservation, detection in human systems may require adjustment of antibody dilution and incubation conditions.
