Biological Background: YME1L1 Function and Localization
- ATP-dependent zinc metalloprotease YME1L1 (alias FTSH1, MEG4, PAMP) is a member of the AAA ATPase family and functions primarily in the mitochondrial inner membrane.
- YME1L1 catalyzes the degradation of both folded and unfolded proteins with degron sequences in the mitochondrial intermembrane region, playing a key role in mitochondrial protein quality control.
Related references: PMID:24315374, PMID:26923599, PMID:27786171 - It cleaves OPA1 at site S2, generating a short form essential for maintaining normal mitochondrial cristae morphology and protein metabolism.
Related references: PMID:18076378, PMID:26923599, PMID:27495975 - YME1L1 ensures cell proliferation, maintains complex I respiration activity, promotes antiapoptotic activity, and protects mitochondria from accumulating oxidatively damaged membrane proteins.
Related references: PMID:22262461 - In response to nutrient starvation or stress, YME1L1 mediates mitochondrial proteome rewiring by degrading translocases, lipid transfer proteins, and metabolic enzymes, regulated by mTORC1 and LPIN1-mediated phosphatidylethanolamine levels.
Related references: PMID:31695197 - Subcellular localization is the mitochondrion inner membrane, with high expression in cardiac and skeletal muscle mitochondria.
- Additional substrates of YME1L1 include OMA1 (degraded upon membrane depolarization), PRELID1, TIMM17A, MICU1, and STARD7, highlighting its broad role in mitochondrial homeostasis.

Experimental Guidance and Technical Tips
- The immunogen sequence (aa 580-773) lies within a conserved region of the AAA ATPase domain; this may contribute to specific target recognition.
- For western blotting, the predicted molecular weight is approximately 86kDa; validate in relevant mitochondrial fractions or whole-cell lysates.
- For immunofluorescence/immunocytochemistry, mitochondrial inner membrane localization suggests a punctate staining pattern; consider using mitochondrial counterstains (e.g., MitoTracker) for co-localization.
- For ELISA applications, ensure optimal antigen presentation; the antibody is applicable to human samples; cross-reactivity with other species should be validated if required.
