Biological Background: YKL-40/CHI3L1 Function and Localization

  • YKL-40/CHI3L1 is a 39-43 kDa secreted glycoprotein belonging to the glycosyl hydrolase 18 family, which binds chitin with high affinity but lacks chitinase catalytic activity.
  • It plays a pivotal role in tissue remodeling and helps cells respond to and cope with environmental changes, especially in chronic inflammation and repair.
  • Functions as a key mediator in T-helper cell type 2 (Th2) inflammatory response and IL-13-induced inflammation, regulating allergen sensitization, inflammatory cell apoptosis, dendritic cell accumulation, and M2 macrophage differentiation.
  • Facilitates invasion of pathogenic enteric bacteria into the colonic mucosa and lymphoid organs by activating AKT1 signaling and subsequent IL-8 production in colonic epithelial cells.
  • In the lung, it contributes to macrophage-mediated bacterial killing, controls bacterial dissemination, and augments host tolerance against infections; also regulates hyperoxia-induced injury, inflammation, and epithelial apoptosis.
  • Subcellularly, it is secreted into the extracellular space, but is also found in the cytoplasm, perinuclear region, and endoplasmic reticulum.
  • Expression is highly restricted to activated macrophages, articular chondrocytes, synovial cells, and liver, with very low or undetectable levels in non-inflammatory colon, muscle tissues, lung, pancreas, mononuclear cells, or fibroblasts.
  • Post-translational modifications include glycosylation and disulfide bond formation; associated keywords and disease links encompass antimicrobial, apoptosis, asthma, and schizophrenia.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to a C-terminal peptide (aa 284–383), which may recognize both full-length and cleaved forms of YKL-40. For Western blot, a band around 43 kDa is expected, but consider that glycosylation can cause slight mobility shifts.
  • Validate the antibody in positive control samples known to express YKL-40, such as activated macrophages, inflamed synovium, or liver tissue. Include appropriate negative controls (e.g., normal colon or fibroblasts) where expression is low.
  • For immunofluorescence applications, note that the protein can localize to the cytoplasm, perinuclear area, and secretory vesicles; permeabilization is required for intracellular detection.
  • In ELISA, pairing this antibody with a capture/detection partner requires in-house optimization; test against recombinant YKL-40 or natural protein in conditioned media.
  • Cross-reactivity with mouse and rat is reported, but signal intensity may vary. Verify species-specific reactivity and titrate antibody concentrations accordingly for each application.
