Biological Background: YKL-40/CHI3L1 Function and Localization

• Chitinase-3-like protein 1 (CHI3L1), also known as YKL-40, BRP39, GP-39, and CGP-39, is a secreted glycoprotein belonging to the glycosyl hydrolase 18 family. It lacks chitinase activity but retains chitin-binding lectin properties.
• Plays a central role in tissue remodeling and cellular responses to environmental changes. It is involved in modulating the extracellular matrix and cell-matrix interactions.
• Functions as a key regulator of T-helper cell type 2 (Th2) inflammatory responses and IL-13-induced inflammation. It influences allergen sensitization, inflammatory cell apoptosis, dendritic cell accumulation, and M2 macrophage differentiation.
• In the colon, CHI3L1 facilitates the invasion of pathogenic enteric bacteria into the mucosa and lymphoid organs, and mediates AKT1 signaling pathway activation and subsequent IL-8 production in colonic epithelial cells.

• The immunogen corresponds to residues 30-389 of mouse YKL-40 (mature protein), suggesting the antibody recognizes the secreted form. Cross-reactivity with human is reported; validation in the relevant species and tissue system is recommended.
• For Western blotting, a band near 43 kDa is expected. Total protein lysates from mouse lung or inflamed colon tissues are suitable positive controls. Optimize blocking and antibody dilution for sample type.
• For IHC-P, use paraffin-embedded sections of mouse lung or mammary tumor as positive tissue. Antigen retrieval may be required; validate with an isotype control.
• For ELISA, the polyclonal format allows both capture and detection roles. Pair with a well-characterized monoclonal anti-YKL-40 to develop a sandwich ELISA, and validate with recombinant protein standards.
