Biological Background: YB-1/YBX1 Function and Localization
- YB-1/YBX1 is a multifunctional DNA- and RNA-binding protein implicated in translational repression, mRNA stabilization, splicing, DNA repair, and transcriptional regulation. Related references: PMID:10817758, PMID:11698476, PMID:14718551
- As an RNA-binding protein, YB-1 preferentially recognizes 5'-[CU]CUGCG-3' motifs and m5C-modified transcripts, and promotes mRNA stability by recruiting ELAVL1. Related references: PMID:10817758, PMID:11698476, PMID:19561594, PMID:31358969
- YB-1 plays a key role in exosome RNA sorting, directing small non-coding RNAs (tRNAs, Y RNAs, miRNAs) into exosomes by recognizing m5C-modified RNAs. Related references: PMID:27559612, PMID:29073095, PMID:28341602
- It regulates alternative pre-mRNA splicing by binding to splice sites and modulating splice site selection. Related references: PMID:12604611
- YB-1 also binds DNA at Y-box sequences (e.g., in MDR1 and HLA class II promoters) to regulate transcription, and has endonucleolytic activity possibly involved in DNA repair. Related references: PMID:8188694, PMID:18809583, PMID:14718551
- Subcellularly, YB-1 localizes to the cytoplasm, nucleus, cytoplasmic granules, P-bodies, and is secreted via exosomes; it can act as an extracellular mitogen. Post-translational modifications include acetylation, phosphorylation, and ubiquitination. Related references: PMID:19483673
Experimental Guidance and Technical Tips
- The immunogen (amino acids 1-55) covers the N-terminal region of human YB-1, which is highly conserved across human, mouse, and rat, supporting the observed cross reactivity.
- For Western blotting, a band around 36 kDa is expected; verify expression in relevant cell lines or tissues.
- In IF/ICC, permeabilization is necessary for nuclear detection; consider co-staining with organelle markers.
- For ELISA, the antibody may be used with recombinant YB-1 or cell lysates; optimize coating and detection conditions.
