Biological Background: YAP1 Function and Localization

Experimental Guidance and Technical Tips
- The immunogen (residues 155–504) covers multiple regulatory domains including TEAD-binding motifs and portions of the transactivation domain; the antibody may recognize full-length YAP1 and some splice isoforms.
- For Western blot, a predominant band near 54 kDa is expected; however, additional bands may appear due to phosphorylation, ubiquitination, or isoform variation.
- Validate antibody specificity in your model system using siRNA knockdown or Hippo pathway modulators (e.g., serum starvation to induce YAP1 phosphorylation and cytoplasmic retention).
- For immunofluorescence, note that YAP1 localization is cell-density dependent; under sparse conditions it is nuclear, while under high confluence it becomes cytoplasmic.
- Optimize antigen retrieval for IHC-P and include appropriate controls (e.g., tissue known to overexpress YAP1, such as certain carcinomas).
