Biological Background: XPOT Function and Localization

  • Exportin-T (XPOT), also designated XPO3 or tRNA exportin, is encoded by the XPOT gene and functions as a specific export receptor for aminoacylated tRNAs.
  • In the nucleus, XPOT binds mature aminoacylated tRNAs and the GTP-bound form of Ran (Ran-GTP) to form a trimeric nuclear export complex.
  • The export complex docks at the nuclear pore complex (NPC) via interactions with nucleoporins, facilitating translocation to the cytoplasm.
  • Once in the cytoplasm, Ran-GTP hydrolysis (stimulated by RANBP1 and RANGAP1) triggers complex disassembly and release of the tRNA cargo; XPOT then recycles back to the nucleus.
  • Directional transport is maintained by the asymmetric distribution of Ran-GTP (high in nucleus) and Ran-GDP (high in cytoplasm).
  • XPOT is predominantly localized to the nucleus and cytoplasm and undergoes shuttling between these compartments during export cycles.
  • Post-translational modifications include acetylation, and the protein has been identified by direct protein sequencing and proteomics.
  • Keywords associated with XPOT include RNA-binding, tRNA-binding, transport, nucleus, cytoplasm, acetylation, and reference proteome.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal region (amino acids 1-200) of human XPOT; consider that epitope availability may vary in certain experimental conditions.
  • This polyclonal antibody is suitable for Western blot (WB) and ELISA; for WB, a band around 110 kDa is expected, but validate in the relevant species and sample system.
  • Cross-reactivity is reported for human, mouse, and rat; ensure sequence homology for other species if needed.
  • For optimal results, test antibody concentration and blocking conditions in your specific assay due to the dynamic localization of XPOT.
