Biological Background: XPNPEP3 Function and Localization
- Catalyzes the removal of a penultimate prolyl residue from the N-termini of peptides, such as Leu-Pro-Ala. Also shows low activity towards peptides with Ala or Ser at the P1 position. Related references: PMID:25609706 PMID:28476889
- Promotes TNFRSF1B-mediated phosphorylation of MAPK8/JNK1 and MAPK9/JNK2, suggesting an adaptor protein function independent of peptidase activity. May inhibit apoptotic cell death induced via TNF-TNFRSF1B signaling.
- Localizes to the mitochondrion and cytoplasm.
- Widely expressed, with isoform 1 being more abundant than isoform 2.
- Belongs to the M24 metallopeptidase family; requires manganese as a cofactor.
- Contains a mitochondrial transit peptide; associated with nephronophthisis (disease variant).
- Alternative splicing produces multiple isoforms, with isoform 1 representing the canonical sequence.

- The immunogen corresponds to the N-terminal region (amino acids 1–250) of human XPNPEP3; confirm epitope conservation in predicted cross-reactive species.
- For Western blot, a primary band near 57kDa is expected, but additional bands may reflect isoforms or post-translational modifications; validate using knockout controls or relevant cell lysates.
- In IF/ICC, the mitochondrial and cytoplasmic localization should be observed; consider co-staining with organelle-specific markers to confirm.
