Biological Background: XPNPEP3 Function and Localization

  • XPNPEP3 (Xaa-Pro aminopeptidase 3), also known as Aminopeptidase P3 (APP3), is a manganese-dependent metalloprotease belonging to the M24B family of aminopeptidases. It is encoded by the XPNPEP3 gene and undergoes alternative splicing to generate multiple isoforms.
  • The enzyme catalyzes the removal of a penultimate prolyl residue from the N-termini of peptides, with preference for Pro, but also shows low activity towards peptides with Ala or Ser at the P1 position. Related references: PMID:25609706 PMID:28476889
  • Beyond its catalytic role, XPNPEP3 functions as an adaptor protein in TNF signaling: it promotes TNFRSF1B-mediated phosphorylation of MAPK8/JNK1 and MAPK9/JNK2, independently of its peptidase activity, and may inhibit TNF-induced apoptotic cell death.
  • Subcellular localization studies reveal that XPNPEP3 is primarily targeted to the mitochondria and is also found in the cytoplasm; the presence of a mitochondrial transit peptide facilitates its import.
  • XPNPEP3 is widely expressed across tissues, with isoform 1 being the more abundant transcript.
  • Mutations in XPNPEP3 have been associated with nephronophthisis-like nephropathy (NPHPL1), highlighting its importance in renal function.
  • The protein contains a 3D structure and features critical metal-binding residues essential for its catalytic activity.

Experimental Guidance and Technical Tips

  • The immunogen comprises amino acids 32–230 of human XPNPEP3 (NP_071381.1), a region N-terminal to the catalytic domain; thus the antibody may recognize multiple isoforms and proteolytic fragments.
  • In Western blotting (WB), multiple bands corresponding to the observed molecular weights (31 kDa, 32 kDa, 48 kDa, 54 kDa, 57 kDa) may be detected; validate the expected bands using positive controls (e.g., human or mouse cell lysates expressing XPNPEP3) and consider the isoform pattern in your sample system.
  • For ELISA applications, the polyclonal nature of this antibody can provide robust signal; however, appropriate blocking and washing steps should be optimized, and assay performance should be verified with recombinant protein or native antigen standards.
  • Given the antibody’s cross-reactivity with mouse XPNPEP3, it is suitable for studies in mouse models; confirm reactivity in your specific tissue or cell type.
