Biological Background: XPNPEP2 Function and Localization
- XPNPEP2 encodes Xaa-Pro aminopeptidase 2, also known as membrane-bound aminopeptidase P (mAmP) or aminoacylproline aminopeptidase, a member of the M24 metallopeptidase family.
- The enzyme functions as a membrane-bound zinc-dependent metalloprotease that specifically removes N-terminal penultimate proline residues from peptides, such as Arg-Pro-Pro.
- It plays a key role in the metabolism of the vasodilator bradykinin, potentially modulating vascular tone and blood pressure regulation.
- The protein is localized to the cell membrane and anchored via a glycosylphosphatidylinositol (GPI) moiety, as indicated by its GPI-anchor and cell membrane keywords.
- XPNPEP2 is glycosylated and contains a signal peptide, with a predicted molecular weight of approximately 76 kDa (75625 Da per UniProt).
- Tissue expression is predominantly observed in kidney, lung, heart, placenta, liver, small intestine, and colon, while absent in brain, skeletal muscle, pancreas, spleen, thymus, prostate, testis, and ovary.
Experimental Guidance and Technical Tips
- The immunogen comprises amino acids 22–240 of human XPNPEP2 (NP_003390.4), corresponding to the N-terminal ectodomain; the antibody is therefore expected to recognize the extracellular portion of the protein.
- For western blotting (WB), a band at ~76 kDa is expected; note that glycosylation may cause the protein to migrate at a slightly higher molecular weight.
- Cross-reactivity with mouse and rat orthologs allows the antibody to be applied in common rodent model studies; appropriate positive and negative tissue controls should be included.
- In ELISA applications, the polyclonal nature may provide broad epitope coverage; consider using it in capture or detection formats and validate performance with recombinant protein standards.
