Biological Background: XPD/ERCC2 Function and Localization
- XPD/ERCC2 is an ATP-dependent 5'-3' DNA helicase and a core component of the transcription/DNA repair factor TFIIH complex, essential for transcription initiation and nucleotide excision repair (NER). Related references: PMID:31253769 PMID:8413672 PMID:9771713
- In NER, XPD/ERCC2 recognizes damaged bases, unwinds DNA around lesions, and facilitates lesion verification and excision of damaged oligonucleotides. Related references: PMID:17466626 PMID:23352696 PMID:31253769
- In transcription, XPD/ERCC2 bridges the CAK kinase module to the core TFIIH complex, enabling promoter opening and RNA polymerase II CTD phosphorylation. Related references: PMID:10024882 PMID:17466626 PMID:33902107
- XPD/ERCC2 localizes primarily to the nucleus, but also associates with the cytoplasm and mitotic spindle, where it participates in chromosome segregation as part of the MMXD complex.
- The protein is also known as ERCC2, BTF2 p80, TFIIH p80, and CXPD; mutations in the ERCC2 gene are linked to Xeroderma pigmentosum, Cockayne syndrome, and trichothiodystrophy.
- XPD/ERCC2 contains iron-sulfur clusters and an ATP-binding domain, and its activity is regulated by post-translational modifications such as ubiquitination.
Experimental Guidance and Technical Tips
- For WB, the predicted molecular weight is approximately 87 kDa; use whole-cell lysates from human or rat cells and consider appropriate loading controls.
- For IF/ICC, validate the antibody in your specific cell/tissue system, as fixation and permeabilization conditions may affect signal.
- As a polyclonal antibody, batch-to-batch consistency should be monitored; pre-adsorption against the immunogen can help confirm specificity.
- This antibody is suitable for ELISA; optimization of coating antigen and detection conditions is recommended.
