Biological Background: XPD/ERCC2 Function and Localization

  • The XPD/ERCC2 protein is an ATP-dependent 5'-3' DNA helicase that is a core component of the general transcription and DNA repair factor IIH (TFIIH) complex, playing a critical role in nucleotide excision repair (NER) and transcription initiation. Related references: PMID:31253769, PMID:8413672, PMID:9771713
  • In transcription-coupled NER, XPD/ERCC2 recognizes damaged DNA bases and is sequestered in chromatin on UV-damaged DNA; the ATP-dependent helicase activity is essential for DNA opening around the lesion. Related references: PMID:17466626, PMID:23352696, PMID:9771713
  • During transcription initiation by RNA polymerase II, XPD/ERCC2 acts as a structural bridge between the CDK-activating kinase (CAK) module and the core-TFIIH complex, and its helicase activity is required for promoter opening and escape; large conformational changes occur between NER and transcription complexes. Related references: PMID:10024882, PMID:31253769, PMID:33902107, PMID:17466626
  • Beyond its roles in DNA repair and transcription, XPD/ERCC2 participates in chromosome segregation as part of the mitotic spindle-associated MMXD complex, and it regulates vitamin-D receptor activity.
  • Subcellular localization includes the nucleus, cytoplasm, and cytoskeleton (spindle), consistent with its roles in transcription, DNA repair, and mitosis.
  • XPD/ERCC2 contains a 4Fe-4S iron-sulfur cluster and an ATP-binding domain; mutations are associated with Xeroderma pigmentosum, Cockayne syndrome, and trichothiodystrophy phenotypes.
  • Alternative splicing yields multiple isoforms, and the protein is subject to ubiquitin conjugation.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to a sequence within amino acids 600-700 of human XPD/ERCC2; consider that this region may be conserved across species, contributing to the observed cross-reactivity with mouse and rat.
  • For Western blotting, a band at approximately 87kDa is expected; validate the specific band by comparing with known molecular weight markers and using appropriate controls.
  • In immunohistochemistry on paraffin-embedded sections (IHC-P), antigen retrieval protocols may be necessary for optimal detection; pilot testing with control tissues is recommended.
  • For ELISA, the antibody may be used for capturing or detecting XPD/ERCC2 peptides or recombinant protein; optimization of coating concentration and detection format is advised.
