Biological Background: XPD/ERCC2 Function and Localization
- The XPD/ERCC2 protein is an ATP-dependent 5'-3' DNA helicase that is a core component of the general transcription and DNA repair factor IIH (TFIIH) complex, playing a critical role in nucleotide excision repair (NER) and transcription initiation. - In transcription-coupled NER, XPD/ERCC2 recognizes damaged DNA bases and is sequestered in chromatin on UV-damaged DNA; the ATP-dependent helicase activity is essential for DNA opening around the lesion. Related references: PMID:17466626, PMID:23352696, PMID:9771713
