Biological Background: XPC Function and Localization
- XPC, also known as Xeroderma pigmentosum group C-complementing protein or p125, is a core component of the global genome nucleotide excision repair (GG-NER) pathway.
- It acts as a damage-sensing and DNA-binding factor within the XPC complex, preferentially recognizing distorted DNA structures such as single-stranded loops, mismatched bubbles, and single-stranded overhangs.
Related references:
PMID:10734143, PMID:10873465, PMID:12509299 - XPC forms a functional dimer with RAD23B, which stimulates its DNA repair activity and enhances binding to cisplatin- and UV-damaged DNA.
Related references:
PMID:20028083 - The XPC complex recruits the TFIIH complex, which then scans for lesions in the opposite DNA strand in a 5'-to-3' direction, leading to productive NER.
Related references:
PMID:12547395 - Beyond DNA repair, XPC also functions as a transcription coactivator by interacting with E2F1 and recruiting KAT2A to promote histone H2A.Z acetylation at target gene promoters.
Related references:
PMID:29973595, PMID:31527837 - Subcellular localization includes nucleus, chromosomes, and cytoplasm.
- Keywords highlight its involvement in DNA repair, transcription regulation, disease variant (Xeroderma pigmentosum), and phosphorylation.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 700-940 of human XPC (NP_004619.3). When using this antibody in Western blotting, a band around 106 kDa is anticipated.
- For IHC-P, nuclear and cytoplasmic staining may be observed; include appropriate subcellular localization controls.
- This polyclonal antibody may be suitable for immunoprecipitation; optimization of antibody concentration and incubation conditions is advised.
- In ELISA, consider its use for quantitative detection of XPC, with appropriate standard curves and validation in the relevant sample system.
