Biological Background: XIAP Function and Localization

  • XIAP (X-linked inhibitor of apoptosis), also known as BIRC4, IAP3, or hILP, is a member of the inhibitor of apoptosis (IAP) family, characterized by three baculoviral IAP repeat (BIR) domains and a C-terminal RING-type zinc finger domain.

Experimental Guidance and Technical Tips

  • The immunogen (amino acids 1-200 of human XIAP) covers the N-terminal region containing the BIR domains. This region is critical for XIAP's interaction with caspases and other binding partners; therefore, the antibody may recognize epitopes essential for functional studies.
  • For Western blot, a band at approximately 57 kDa is expected based on the predicted molecular weight of XIAP. However, due to potential post-translational modifications (e.g., phosphorylation, ubiquitination) or cleavage products, additional bands may be observed; validate in your sample system.
  • This antibody has been reported to react with human, mouse, and rat XIAP by the manufacturer. For cross-species applications (e.g., mouse or rat samples), verify reactivity and specificity under your experimental conditions.
  • The antibody is KO validated, indicating a high probability of specificity. Still, include appropriate positive and negative controls (e.g., XIAP knockout lysates) in your assays.
  • For ELISA, optimize coating antigen concentration and antibody dilution; the immunogen is a recombinant protein, so consider using a similar recombinant protein as a positive control.
