Biological Background: WWOX Function and Localization
- Protein Identity and Isoforms: WWOX encodes the WW domain-containing oxidoreductase, also known as FOR, WOX1, or SDR41C1. Alternative splicing produces multiple isoforms; isoform 5 and isoform 6 may be restricted to tumor cell lines.
- Tumor Suppressor and Apoptosis: Acts as a tumor suppressor and plays a role in apoptosis. Required for normal bone development (by similarity). May function synergistically with p53/TP53 to control genotoxic stress-induced cell death.
- TGFB1 and TNF Signaling: Plays a role in TGFB1 signaling and TGFB1-mediated cell death. May also participate in tumor necrosis factor (TNF)-mediated cell death.
- Wnt Pathway Inhibition: Inhibits Wnt signaling, probably by sequestering DVL2 in the cytoplasm.
- Subcellular Localization: Localizes to the cytoplasm, nucleus, mitochondrion, Golgi apparatus, and lysosome.
- Tissue Expression: Widely expressed, with strong expression in testis, prostate, and ovary. Overexpressed in some cancer cell lines.
- Post-Translational Modifications and Keywords: Annotated as a phosphoprotein and subject to ubiquitin conjugation. Keywords include alternative splicing, disease variant (epilepsy, neurodegeneration), and NADP binding.

- The immunogen corresponds to the N-terminal region (aa 1-120) of human WWOX, which may detect full-length and some N-terminal isoforms. For Western blot, consider a validated positive control lysate from a cell line with endogenous expression.
- As a rabbit polyclonal antibody, the product exhibits broad reactivity (human, mouse, rat). Optimize the dilution factor for each application; typical starting dilutions for WB are 1:500-1:2000.
