Biological Background: WTAP Function and Localization

  • WTAP (Wilms tumor 1-associating protein), also known as Mum2 or hFL(2)D, is a pre-mRNA-splicing regulator and a core component of the WMM complex.
  • As a component of the WMM complex, WTAP mediates N6-methyladenosine (m6A) methylation of RNAs, influencing mRNA splicing and processing efficiency.   • WTAP functions as a mRNA splicing regulator, influencing the processing of numerous transcripts. Related references: PMID:12444081

  • The immunogen covers amino acids 212-355 in the central region of human WTAP; the antibody may recognize multiple WTAP isoforms if the epitope is conserved.
  • For Western blotting, expect a band near the predicted molecular weight of 44 kDa. Validate in appropriate cell lines or tissue lysates expressing WTAP.
  • In immunofluorescence and immunocytochemistry, WTAP should exhibit a punctate nuclear speckled staining pattern, consistent with its known localization.
  • Cross-reactivity with mouse and rat WTAP enables studies across species; however, confirm specificity in your experimental system using appropriate controls.
  • For ELISA, titrate the antibody and validate using recombinant WTAP protein or peptide to ensure optimal signal-to-noise ratio.
