Biological Background: WNT5A Function and Localization
- Protein Wnt-5a, also known as WNT5A, is a member of the Wnt family of secreted signaling proteins.
- Wnt-5a acts as a ligand for Frizzled receptors, capable of both activating and inhibiting canonical Wnt signaling depending on receptor context. With FZD4 it activates beta-catenin signaling, while with ROR2 it inhibits the canonical pathway by promoting beta-catenin degradation through a GSK3-independent mechanism (By similarity).
- Essential during embryogenesis for extension of the primary anterior-posterior axis, outgrowth of limbs and genital tubercle, and inhibition of type II collagen expression in chondrocytes, facilitating chondrogenesis (By similarity).
- Functions as a tumor suppressor by suppressing proliferation, migration, invasiveness, and clonogenicity of carcinoma cells. Related references: PMID:15735754

- Secreted into the extracellular space and extracellular matrix.

Experimental Guidance and Technical Tips
- For Western blotting, a band around 42 kDa is expected; however, post-translational modifications such as glycosylation and palmitoylation may result in a higher observed molecular weight.
- For immunohistochemistry (IHC-P), consider antigen retrieval methods like heat-induced epitope retrieval to enhance detection.
- For immunofluorescence (IF/ICC), permeabilization is required to detect the secreted protein, which may also localize intracellularly.
- The immunogen corresponds to amino acids 116–215; this peptide can be used as a blocking control in ELISA or for epitope competition assays.
