Biological Background: WNT4 Function and Localization
- WNT4 encodes a secreted glycoprotein, Wnt-4, which acts as a ligand for frizzled family receptors. This protein is involved in the Wnt signaling pathway and undergoes alternative splicing, producing multiple isoforms.
- Wnt-4 plays a critical role in embryonic urogenital development. It is required for normal mesenchyme-to-epithelium transition during kidney development and for the formation of early epithelial renal vesicles (By similarity). In females, it is essential for the formation of the Müllerian duct and normal oocyte levels in the ovaries, and it regulates the down-regulation of 3β-hydroxysteroid dehydrogenase. - Wnt-4 is required for normal lung development and proper patterning of tracheal cartilage rings (By similarity).
- Wnt-4 is secreted into the extracellular space and associates with the extracellular matrix, consistent with its role as a signaling molecule.
- As a Wnt protein, Wnt-4 is predicted to contain conserved disulfide bonds and undergo lipidation (palmitoylation) and glycosylation, which are essential for its secretion and signaling activity.
- Mutations in WNT4 are linked to SERKAL syndrome and other developmental disorders, highlighting its importance in human disease.
Experimental Guidance and Technical Tips
- This polyclonal antibody is raised against a recombinant fragment spanning amino acids 122–351 of human WNT4, likely recognizing multiple epitopes. Consider using it in Western blot under reducing and non-reducing conditions to detect endogenous WNT4 (predicted molecular weight ~39 kDa).
- For ELISA applications, pairing with a suitable detection antibody may be necessary. Validate the assay in the relevant sample matrix (e.g., tissue lysates or cell supernatants).
- Given cross-reactivity with mouse and rat, this antibody is suitable for cross-species studies in developmental models. Note that Wnt-4 is a secreted protein, so sample preparation should focus on tissue extracts or conditioned media.
- Include appropriate controls (e.g., knockout cell lines or siRNA-treated samples) to confirm specificity, as Wnt family proteins share conserved regions.
